Hexarelin (also designated as Examorelin) is a synthetic hexapeptide derived from GHRP-6 that functions as a potent, selective growth hormone secretagogue receptor (GHSR) agonist. Mechanism of Action
Hexarelin exerts its primary endocrine function by binding to the growth hormone secretagogue receptor type 1a (GHSR-1a) in the pituitary and hypothalamus. This interaction activates the phospholipase C (PLC) and inositol triphosphate (IP3)/diacylglycerol (DAG) intracellular cascade, leading to the mobilization of intracellular calcium stores and the exocytosis of growth hormone vesicles without depleting pituitary reserves. Concurrently, Hexarelin binds to the scavenger receptor CD36 found abundantly in cardiac and vascular tissues. Through CD36 activation, Hexarelin modulates downstream pro-survival kinases, reduces cellular apoptosis, mitigates oxidative stress, and prevents ischemia-reperfusion injury in isolated myocardial research systems.
